چکیده

T HE PROCESS of cellular uptake and metabolism of cyclosporine (CsA) is poorly understood. Previously, we have shown that the transport of CsA in blood occurs in association with plasma Iipoproteins,l with low-density lipoprotein (LOL), and with highdensity lipoprotein, each transporting about 32% of the total amount of CsA in blood. This observation raises the possibility that further cellular metabolism of CsA, at least in part, may take place in conjunction with LOL metabolism through its specific, high-affinity cell surface receptor. In this report, we present results of studies that indicate that (1) [lH]_ labeled CsA is taken up, in an apparently specific manner, by peripheral blood lymphocytes (PBL) when incubated with the [lH] CsA-LOL complex; and (2) cellular uptake of [lH] CsA is enhanced when PBL, whether freshly isolated or stimulated with phytohemagglutinin (PHA), are incubated in a medium containing both [lH] CsA and FK506 (FK) in a 1:1 (wt/wt) proportion.

Inhibition of tumour necrosis factor and IL-17 production by leflunomide involves the JAK/STAT pathway

OBJECTIVE To study the effects of different disease-modifying antirheumatic drugs (DMARD) on different events mediated by IL-15-activated lymphocytes. METHODS Peripheral blood lymphocytes (PBL) were isolated from healthy donors and activated with IL-15 after exposure to different DMARD: leflunomide, cyclosporin A, methotrexate, mycophenolic acid, FK-506, sulphasalazine and sodium aurothiomala...

Serial monitoring of immunologic function and phenotype of lymphocytes in the blood of transplanted patients randomized to cyclosporine or FK 506.

EFFECTS of the drugs FK 506 and CyA on lymphocyte function have been mostly studied in vitro and in vivo in animal models. 1-5 Little information is available about changes in the immune system of patients who, having undergone orthotopic liver transplantation (OL T), are treated with immunosuppressive drugs. To evaluate cellular immunity of patients treated for prolonged periods with FK 506 or...

Pretransplantation conditioning influence on the occurrence of cyclosporine or FK-506 neurotoxicity in allogeneic bone marrow transplantation.

BACKGROUND AND PURPOSE Transplantation conditioning regimens have been shown to affect the brain imaging appearance in patients with cyclosporine or FK-506 neurotoxicity. We assessed whether the occurrence of neurotoxicity was affected by the choice of conditioning regimen used before allogeneic bone marrow transplantation (allo-BMT). METHODS An allo-BMT was performed in 290 patients conditio...

Immunosuppressive effect of FK-506 on in vitro lymphocyte alloactivation: synergism with cyclosporine A.

R ECENT STUDIES have described a new potent immunosuppressive drug FK-506 (FK) that induces considerable prolongation of allograft survival.'·) This drug is isolated from Streptomyces tsukubaensis. has a molecular weight of 822 daltons, and is structurally different from cyclosporine A (CsA). In vitro investigations have demonstrated a strong inhibitory effect on lymphocyte activation that appe...

Correlation between bioassayed plasma levels of FK 506 and lymphocyte growth from liver transplant biopsies with histological evidence of rejection.

FK 506, a novel immunosuppressive agent, was first used to reverse rejection in liver recipients who had failed to respond to conventional therapy.' The successful outcome of the initial study has led to clinical trials of FK 506 as the primary immunosuppressive agent for liver allografts and other organ transplants. 2-4 Ongoing studies include dosage protocols for optimal immunosuppression wit...
